| 胃癌的高死亡率与其转移和化疗耐药性密切相关。铁死亡作为一种新型铁依赖性程序性细胞死亡方式,在胃癌进展中发挥重要作用。目前研究表明,谷胱甘肽过氧化物酶4-谷胱甘肽(GPX4-GSH)抗氧化系统失衡是胃癌细胞铁死亡的核心机制。肠道菌群能通过微生物代谢产物、细胞因子调节以及铁离子稳态维持等途径,影响胃癌细胞的铁死亡进程,这一作用在幽门螺杆菌(Hp)根除后的肠道菌群重建过程中表现得尤为显著。此外,多种天然化合物具有通过诱导铁死亡增强化疗敏感性的潜力。本文提出了多组学整合分析构建铁死亡调控网络,阐明微生物-宿主互作机制,并探讨基于微生物组修饰结合铁死亡干预的个体化治疗策略,系统阐述肠道菌群、铁死亡与胃癌三者间的分子互作网络和临床应用前景。 |

Gut Microbiota-regulated Ferroptosis in Gastric Cancer: Mechanisms and Therapeutic Prospects

| The high mortality of gastric cancer is closely linked to tumor metastasis and chemoresistance. Ferroptosis, an novel iron-dependent form of programmed cell death, plays a pivotal role in gastric cancer progression. Studies reveal that the imbalance of the glutathione peroxidase 4-glutathione (GPX4-GSH) antioxidant system is the core mechanism of ferroptosis in gastric cancer cells. Gut microbiota can influence the ferroptosis process in gastric cancer cells through pathways such as microbial metabolites, cytokine modulation, and iron ion homeostasis maintenance. This effect is particularly pronounced during the gut microbiota restoration process following Helicobacter pylori (Hp) eradication. Additionally, various natural compounds have the potential to enhance sensitivity to chemotherapy by inducing ferroptosis. This article proposed integrating multi-omics analysis to construct ferroptosis regulatory networks, elucidating molecular mechanisms of microbe-host interactions, and exploring personalized therapeutic strategies based on microbiome modification combined with ferroptosis intervention. The molecular interaction network among gut microbiota, ferroptosis, and gastric cancer, as well as its clinical application prospects were systematically explored. |
